An Electron Microscope Study of Isolated Mitochondria

چکیده
1. The present paper constitutes a preliminary study of the morphology of mitochondria by means of electron microscopy. 2. The mitochondria that were the subject of this investigation were obtained from a lymphosarcoma of the rat. They were separated from the other components of the leukemic cells by a method of differential centrifugation, and thus made available for direct examination in the electron microscope. 3. In the purified form the mitochondria appeared as spherical bodies, the majority of them varying in size approximately from 0.6 to 1.3 micro in diameter. 4. Certain aspects of mitochondria in the electron microscope suggest that these elements are surrounded by a differentiated membrane. In some cases the limiting membrane seemed to be responsible for maintaining the general shape of the mitochondria, even when most of the mitochondrial substance had been lost. 5. By means of the electron microscope, it is possible to distinguish small elements, 80 to 100 mmicro in diameter, within the body of certain mitochondria. Further work is suggested to establish whether these small granules are normal constituents of mitochondria, and what relation may exist between them and ordinary microsomes. 6. The nature of mitochondria as morphological units is discussed. Present evidence indicates that mitochondria constitute definite physical entities which can persist in the absence of the cytoplasm.

Biochemical and Cytological Changes Accompanying Growth and Differentiation in the Roots of Zea mays
The apical meristem of the root affords an excellent material with which to study changes in cellular components accompanying growth and differentiation. The ontogeny of cytoplasmic particles can be followed, since the younger cells are constantly dividing and reforming new cytoplasm. Electron microscope pictures of these newly formed cells reveal a dense background of microsomal granules and s...
متن کاملIsolation of Highly Purified Mitochondria from Rat Pancreas
The exocrine pancreas, an active secretory gland, is a suitable model for studies concerning the structure of cell organelles and the coupling of energy production with different biosynthetic processes (1, 2) . However, the investigation of isolated pancreas mitochondria is not often performed because of the many difficulties in obtaining reproducible preparations with well-preserved structure ...
متن کاملAn Electron Microscope Study of Rhabdomyosarcoma.
An electron microscope study of idtraihin sections of two rhabdomyosarcomas revealed the presence of cytologic characteristics previously described in neoplasias: nuclear envelope invagination, reduction in number of organelles, reduced size of mitochondria, sioollen mitochondria. In addition, one group of seven centrioles was observed and many cells revealed the presence of aggregates of small...
متن کاملMorphology and Atp-ase of Isolated Mitochondria
Changes in the morphology of rat liver mitochondria brought about by different methods of isolation and the concomitant changes in ATP-ase activity were studied. The morphology was investigated with the electron microscope. It was found that the ATP-ase activity of the isolated mitochondria cannot be readily correlated with the morphology of the mitochondria. The ATP-ase found in these preparat...
متن کاملStudy of Mitochondria in Rat Liver
The electron microscope has been used to determine the weight distribution of isolated subcellular particles from normal rat liver. The following results are reported: (1) There exist at least two well defined weight populations of subcellular particles; their respective median weights are 1.3 x 10(-14) and 11 x 10(-14) gm. The lighter fraction is considered to consist of lysosomes, the heavier...
